Picard and Crusher met while Picard served with Beverly's late husband, Jack Crusher on the Stargazer.
After Jack's death in an accident aboard The Stargazer, Beverly and Jean Luc parted company, not seeing each other again until her arrival to serve on the Enterprise.
Since that time, their relationship has been one of secret feelings for the other, sometimes being made apparent in strange circumstances, such as the time when the entire ship was affected by a intoxicating illness that caused the crew to lose their inhibitions and do things they ordinarily would not have. In an alternate timeline, they have even been married (and divorced). (All Good Things...)
Word has it that the Picard/Crusher relationship will make a major turn in the next Star Trek movie(ST:9), as yet unnamed.

William and Deanna met at a traditional Betazoid wedding on Deanna's home planet of Betazed. While Riker pursued Deanna, she vowed to have nothing to do with him, until she realized that they were indeed meant for each other -- they were Imzadi, meaning "beloved" in a spiritual sense. Over time, however, they drifted apart, only to be reunited again on board the Enterprise-D.
