Melbourne's tram route 5 starts from the Melbourne University terminus of St Kilda Rd services at the corner of Swanston and Elgin Sts Carlton. Route 5 trams share Swanston St and St Kilda Rd with routes 1,3,6,8,16,22,64,67 and 72. Trams pick up their first passengers at the Faraday St stop and then go down Swanston St past the Royal Women's Hospital in Grattan St and the enter Melbourne's CBD at Victoria St opposite the City Baths.
On their way through the city, trams cross LaTrobe St (routes 23,24,30,34 and the City Circle tourist tram), Lonsdale St (National Bus Company routes to the Eastern suburbs), Bourke St ( route 86 to Bundoora RMIT and route 96 between St Kilda Beach and East Brunswick), Collins St ( route 11 to West Preston, route 12 to St Kilda and route 109 between Mont Albert and Port Melbourne) and Flinders St ( route 48 to North Balwyn, route 70 to Wattle Park and route 75 to East Burwood).
Crossing Flinders St, route 5 trams pass Federation Square on the left and Flinders St station on the right then cross Princes Bridge over the Yarra River, then passing the Victorian Arts Centre and National Gallery on the right before Southbank Boulevarde and its junction with route 1 to Sth Melbourne Beach.
Trams now pass through an area populated with high-rise office blocks, some of which have been converted into expensive, trendy, completely unaffordable apartments, although extensive gardens are in evidence on the left hand side of St Kilda Rd as trams proceed past the Shrine of Remembrance and then arrive at the Domain Rd Interchange, born in 1988 courtesy of a Federal Government Bicentennial grant. Route 55 to West Coburg via the Zoo leaves here, journeying down Park St to the right, while route 8 to Toorak - Glenferrie Rd disappears to the left up Domain Rd.
At Commercial Rd, route 72 to Camberwell turns off, as does route 6 to Glen Iris at High St, before the tram arrives at St Kilda Junction, where route 5 turns left.
Route 5 shares track with route 64 to East Brighton along Dandenong Rd before trams arrive at Wattletree Rd Malvern, where route 5 turns off, having previously crossed Chapel St ( route 78 between St Kilda Beach and North Richmond), the Sandringham railway line and Orrong Rd, where at night after 7.00 pm route 5 passengers must board a route 64 tram in the city and change to a route 5 shuttle here at Orrong Rd.
Wattletree Rd is a road lined with houses and small shops, this only being relieved at the Glenferrie Rd crossing of route 69 between St Kilda Beach and Kew - Cotham Rd, which runs through the middle of the Glenferrie Rd shopping centre. Route 5 continues on for a short distance to its terminus at Burke Rd, where there is a single track terminus with a safety zone, seating being provided on the footpath adjacent to the terminus.
