Abstract

Chick and quail embryos ages 24-96 hours were fixed in Bouin's fixatives for at least 24 hours and then stained with Ehrlich acid hematoxylin. The technique was based on Billett and Wild (1975). The result showed purple-red colour. Embryos of later ages were fixed in ethyl acohol and cartilage and bone were pale blue and dark red respectively and muscle was colourless.

บทคัดย่อ

เอมบริโอนกกระทาญี่ปุ่นและไก่ อายุฟัก 24 ชั่วโมง-96 ชั่วโมง ถูกนำมาศึกษาเทคนิคการทำสไลด์ถาวร โดยดัดแปลงมาจากวิธีของ Billett และ Wild (1975) โดยดองในน้ำยาโบแอง (Bouin's fixative) และย้อม ด้วยสี Enrlich acid Hematoxylin ผลปรากฏว่าเอมบริโอติดสีม่วงแดง ส่วนเอมบริโออายุฟักจาก 5 วัน จนถึงฟักเป็นตัว นำมาศึกษาโดยวิธีดองใส ซึ่งดัดแปลงมาจากวิธีการของ Dingerkus และ Uhler (1977) โดยใช้น้ำยาเอทิลอัลกอฮอล์ ดองสัตว์และไม่ใช้เอนไซม์ทริพซีน ผลปรากฏว่าประดูกอ่อนติดสีน้ำเงิน กระดูกแข็งติดสีแดงเข้ม ส่วนกล้ามเนื้อและเนื้อเยื่ออ่อนจะใสไม่ติดสี
