Lane (John Wayne) has taken a job working for Mrs. Lowe. It seems her late husband had stolen a half million dollars in gold and had hidden it in a train engine laying on its side in the dessert in Mexico. Lane sent word to some friends to help him with the job. There was Will Jesse (Ben Johnson), Grady (Rod Taylor), and Ben Young (Bobby Vinton). Grady brought along a couple of his friends, Calhoun (Chris George), and Sam Turner (Jerry Gatlin).

Shortly after they meet and start out for Mexico, several more riders come by train to the town where Lane and his men had left from to follow them with the intent of killing them and taking the gold for themselves. Mrs. Lowe wanted to return the gold to Wells Fargo to clear her husbands name for the sake of their son. The reward for returning the gold would be $50,000.00 that was what Lane was going to get for helping.

They find the gold, battle the men following, killing some and return to the town they started from to put Mrs. Lowe and the gold on the train. There is yet another man following them and the other riders, a Pinkerton Agent (Ricardo Montalban). Lane and his friends decide to let Mrs. Lowe keep the reward money. They tell her as she is boarding the train, she is almost in tears as the train starts pulling away. They start to walk off as they pass the rear of the train, the pinkerton agent is standing on the rear platform. As the train starts to pick up speed he tells Lane that their gesture was very touching but, the woman's real name was Lilly, she was with Matt Lowe when he died, but that Matt Lowe was never married.

Will Jesse and Grady turn to look at Lane but he was gone. In just a few seconds he rides up to them, Will asks him where he's going, and he replies, "To Rob A Train"!!!
