Luis Alfonzo Larrain nació en la ciudad de la Victoria, estado Aragua, Venezuela, el día 22 de Julio de 1911. Es hijo de Ana Larrain Martínez y Alejandro Alfonzo Pérez. En el año 1918 se traslada a Caracas la familia Larrain y reside en la Parroquia La Pastora, en el Centro de la ciudad. Desde muy temprana edad se siente atraído por la música, siendo sus primeras influencias la banda dirigida por el maestro Carlo Bonet y su madre quien le enseña a cantar y tocar el Cuatro. Ya a los nueve años aparece en él, la figura del compositor, que se vio estimulada por la magnifica cantante española Pilar Arcos, quien incluye en su repertorio las composiciones del precoz niño. En 1925, cuando contaba apenas catorce años, Luis formo una pequeña orquesta, junto a los noveles músicos Rafael Briceño (Piano), Rafael Uzcategui (Violín) y Eduardo Serrano (Batería). Luis ejecutaba el Banjo y era el Manager según cuentan ellos mismos. En 1927 la orquesta toma el nombre de Orquesta Flava . En 1928 la orquesta se presentaba en los sitios y eventos más distinguidos de la ciudad, en el Hotel Majestic y en eventos estudiantiles como el Sacalapatalaja, siendo considerado un movimiento artístico antigomecista. Se icorporan a la orquesta Federico Ayesta (Trompeta), Heber Hernandez (Violín), Luis Eladio Guevara (Saxo) y eventualmente el maestro Antonio Lauro (Guitarra).

En 1930 incursiona en la actuación, en la empresa Broadcasting Caracas junto al distinguido empresario William H. Phelps, empresa que con los años se llamaría Radio Caracas. Actúa junto a figuras como Cecilia Martínez, Margot Antillano, Conchita Ascanio, Edgar J. Anzola, Francisco Fossa y Ricardo Espina. Por esta fecha dirige las agrupaciones musicales Los Acróbatas del Pentagrama , Los Hijos de la Noche, El Cuarteto Melódico Luis Alfonso Larrain, en el que participan Graciela Naranjo, Fidel Peters, Ofelia Ramón y Enrique Alvarenga. En 1937 forma la que llegó a ser la Gran Orquesta Luis Alfonzo Larrain que luego le valió el nombre de Mago de la Música Bailable y la Orquesta del Buen Tono . Fue discípulo del maestro Vicente Emilio Sojo y realizo arreglos para los merengues populares criollos que le valieron el estatus aristocrático que permitió su internacionalización y refinamiento.

La unión en matrimonio con Elisa Soteldo, cantante de la orquesta, tuvo como fruto el nacimiento de su hija Keyla Hermecheo, célebre profesional de la Danza Clásica y quien se destaca en el mundo del Ballet mundial. Keyla funda la prestigiosa escuela de Ballet Keyla Hermecheo, semillero para la formación de talentosos bailarines clásicos y también el Ballet Metropolitano de Caracas. Además es codirectora de educación para América de la Alianza Mundial de la Danza en los Estados Unidos de Norteamérica. Keyla Hermecheo fue miembro del Comité Ejecutivo Mundial de la Danza. Para ese tiempo actúan con la Orquesta vocalistas de renombre nacional e internacional tales como Ada Alba, Carmen Chirinos, Flor Díaz, Trino Finol, Luisín Landáez, Marco Tulio Maristani, Kiko Mendive, Manolo Monterrey, Graciela Naranjo, Juanito Renot, Rafa Robles, Elio Rubio, Leo Rodriguez, Hilda Salazar, Mario Suarez, Callos Torres, Orlando Vallejo, Pedro Vargas, Maurice Chevalier. Intervienen los internacionales Carlos Gardel, Tito Guizar, Miguelito Valdez, Teo camargo, Jorge Negrete, Pedro Infante y Nat King Kole. Como compositor tuvo extensa e inspirada producción, siendo el caso que la mayoría de sus composiciones tienen música y letra de él mismo. Algunas de tipo romántico y otras populares.

Paralelamente a su actividad musical, desarrolla actuaciones de carácter intelectual. Mantenía artículos en distintas publicaciones y periódicos sobre Historia del Arte, Temas Musicales y Humor. En 1939 se destaca en cinematografía desarrollando la producción musical para la película Carambola con argumento y dirección de Fini Veracoechea, letra de Rafael Brunicardi y guión de Sara Cabrera. Funda los Estudios Larrain donde se realizan grabaciones y producciones para discos y casetes. Esta empresa es administrada por su tercera esposa Eduvigis Bravo (Vivita) y culmina sus actividades en 1987. El 24 de Mayo de 1955 funda la SACVEN, Sociedad de Autores y Compositores de Venezuela , importante institución, desde el punto de vista de su proyección social, en beneficio de la seguridad económica de todos sus colegas venezolanos. Redactó el proyecto de la ley de autores y compositores y logró que fuera aprobado por el Congreso Nacional como Ley de la República.
